Current studies show that Peyronie's disease (plastic hardening of the penis) is an inflammatory disease of the penis in which the elastic layer (tunica albuginea) of the penis is damaged in the central tissue (corpora cavernosa). As a result, the affected area loses elasticity, and when filled with blood, it does not stretch but instead leads to a curvature of the penis toward the scar. Peyronie's disease is the most common cause of excessive penile curvature.

The process begins with the oxidation of various fats and protein molecules, leading to inflammation of the elastic layer (tunica albuginea) and the corpus cavernosum. This process continues until the collagen fibers are eventually replaced by inelastic fibers, forming the fibrous plaque (hardened nodules) characteristic of this disease.

These plaques can be felt under the skin, usually on top or sometimes on the sides of the penis. Most of them resolve naturally, but sometimes they persist and, over time, cause:

Abnormal curvature of the penis (often upwards));

• Erectile dysfunction and/or pain during erection;

Currently available treatments are not yet entirely satisfactory and mostly only help alleviate the symptoms of Peyronie's disease. If the curvature of the penis persists in the later stages of the disease, when the pain has already subsided, surgery may be necessary to correct it.

However, this treatment is very invasive and can sometimes result in penile shortening of up to 4 cm (1.6 inches).
